Inability of KU60019 to cross the BBB after chronic administration. Healthy C57/Black mice were inoculated i.p. daily during two consecutive weeks (total of 10 injections) with 30 µl/g body weight of (A) vehicle (2.5% ethanol in 0.9% NaCl) or (B) 250 µM KU60019. Mice were then euthanized by CO₂ asphyxiation, the brain removed and whole brain extracts prepared as described in Materials and Methods and¹³. KU60019 content was determined by HPLC/MS as described in Materials and Methods and¹³. KU60019 amounts higher than limit of quantitation (LoQ - 5 nM) were never found in brain tissues. C. As a chromatographic marker, 10 nM KU60019 was run under the same conditions.
