Inability of KU60019 to cross the BBB after chronic administration. Healthy C57/Black mice were inoculated i.p. daily during two consecutive weeks (total of 10 injections) with 30 µl/g body weight of (A) vehicle (2.5% ethanol in 0.9% NaCl) or (B) 250 µM KU60019. Mice were then euthanized by CO2 asphyxiation, the brain removed and whole brain extracts prepared as described in Materials and Methods and13. KU60019 content was determined by HPLC/MS as described in Materials and Methods and13. KU60019 amounts higher than limit of quantitation (LoQ - 5 nM) were never found in brain tissues. C. As a chromatographic marker, 10 nM KU60019 was run under the same conditions.
