Soluble IgG is essential for the BCR targeted in vivo deletion induced by SpA. B-cell deficient (muMT) mice received adoptive transfer of T15i B cells, and after sham or human polyclonal IgG pretreatment were later challenged with PBS or SpA. Results represent residual splenic B220+ IgMa + B cells, after flow cytometric gating on adoptively transferred CFSE-labeled mononuclear cells, which omits CD3+ cells. Treatment with hu IgG alone had no effect (not shown). Gating strategy was used to quantitate values shown in Figures 3, 5.
