Extent of in vivo SpA-mediated B cell depletion reflects in vitro binding affinity. Assessment of SpA mediated depletion of transgenic T15i (+/−) B-cells following sham pretreatment or pretreatment with human polyclonal IgG (hu pIgG) or human polyclonal IgG F(ab)'2 (pIgG Fab'2) or monoclonal murine IgG, as indicated. At 72 h after SpA (or sham) treatment, levels of residual T15i B cells are indicated for individual mice. Results represent residual splenic IgMa + B cells, after gating on adoptively transferred CFSE-labeled mononuclear cells, that identified only the B220+ IgMa+ Cells. After VHIII Fab IgG2a treatment, all mice died acutely (Inline graphic) and residual B cells could therefore not be quantitated.
