Association of dynein and dynactin with capsid-like HIV-1 assemblies in vitro. (A and B) Recombinant CA and CA-NC proteins were assembled into tubes and examined by negative-stain electron microscopy to confirm tubular assembly. CA tubes were incubated with 293T cell extracts. The reactions were then centrifuged, the pellets were washed twice, and the pelleted proteins were analyzed by SDS-PAGE and immunoblotting. Blots were probed with antibodies to dynein heavy chain (C, CA; D, CA-NC), dynein intermediate chain (E, CA; F, CA-NC), and dynactin component ACTR1A (G, CA; H, CA-NC) or CA (C to H). Input samples corresponding to 10% of each reaction were removed prior to pelleting. Representative immunoblots from three independent experiments are shown.
