Similarly to Figure 4, the semi-transparent oval in the first diagram indicates portions of freshly replicated duplexes that will serve as substrate of Topo IV-mediated passages in the analyzed here case. Notice that both indicated portions belong to two different freshly replicated sister duplexes (drawn in red and blue).
