Endogenous DHX9 is required for RNAPII recruitment to NF-κB–dependent antiviral promoters. (A) shCtrl and shDHX9/HeLa cells were infected with MHV-68 at MOI 2 for 12 h. The nuclear extract was subjected to immunoprecipitation with the anti-p65 antibody and immunoblotted with the indicated antibodies. The input lysates represent 10% of nuclear extracts by volume used for each immunoprecipitation. (B–D) shCtrl/HeLa and shDHX9/HeLa cells were infected with MHV-68 at MOI 2 for 12 h. Cell lysates were prepared and analyzed by a ChIP assay with an anti-p65 or anti-RNAPII antibody. Normal rabbit IgG was included as a control for nonspecific immunoprecipitation. The amounts of precipitated DNAs were measured by qPCR using primers flanking the promoter regions of IL6 (B), IFN-β (C), and GAPDH (D). The data represent mean ± SD of triplicate assays.
