Polydatin suppressed hepatocellular lipotoxicity and hepatocyte apoptosis. HepG2 cells were induced by 0.25 mM palmitic acid (PA) in the absence or presence of polydatin (5, 10, 20 μM) for 24 h. (A) Lipid droplets in HepG2 cells were measured by Oil red O staining assay. (B) The contents of triglycerides in HepG2 cells. (C) The mRNA levels of FAS, SREBP-1c were measured by q-PCR. (D) The proportion of LDH release from HepG2 cells. (E) The protein expression of cleaved-caspase3, caspase3 was examined by Western blot and analysis. (F) The apoptotic cells were determined by flow cytometric analysis. #p < 0.05, ##p < 0.01, ###p < 0.001, ####p < 0.0001 vs Control; *p < 0.05, **p < 0.01, ***p < 0.001 vs HepG2 induced by PA.
