PfuHAN nuclease activity is dependent upon the C-terminal DHH nuclease domain. (A) 15% SDS-PAGE of full-length and truncated PfuHANs. Schematic representation of the domain combinations of each protein are listed on the top of each lane in the gel. The number of starting residues for each protein is indicated. The 3′-exonuclease activities of full-length PfuHAN and the five truncated proteins were characterized by incubating enzymes with (B) ssDNA or (C) ssRNA (50 nM) at 55°C for 20 min. The concentration of PfuHAN and its truncated mutants are 0, 10, 20, 50, 100 and 200 nM.
