Preferential organellar accumulation of candidate proteins in transgenic Arabidopsis plants. Leaf epidermal cells of transgenic Arabidopsis thaliana plants constitutively expressing PDF/eYFP (A) or GCS/eYFP (B) show preferential accumulation of the fluorescent reporter protein in plastids and mitochondria, respectively. To compare the relative signal strengths in the two organelles, the degree of fluorescence was quantified using the plot profile tool of Fiji along a single line of 10 μm, covering in each case both a plastid and a mitochondrion. In the resulting graphs, gray value represents fluorescence intensity of the eYFP signal (black line) and the chlorophyll signal (red line). For further details see the legend of Figure 1.
