Nutlin decreases glycolytic genes via MDM2-mediated downregulation of SP1. (A) Cells were treated with Nutlin (10 μM) for 24 h. mRNA was analyzed by qPCR for the indicated genes. (B) Cells were transfected with control siRNA or SP1 RNA for 48 h. mRNA was analyzed by qPCR for the indicated genes. (C) Cells were treated with vehicle or Nutlin (10 μM) for 24 h, or transfected with control siRNA or SP1 RNA for 48 h. mRNA was analyzed by qPCR for TIGAR. Relative mRNA level is presented as graphs with SD indicated (triplicate). (D) Cells were treated with vehicle or Nutlin (10 μM) for 24 h. Lysates were immunoblotted for the indicated proteins. (E) MHM cells were treated with vehicle or Nutlin (10 μM) for 21 h and then additionally with vehicle or MG132 (10 μM) for 3 h. Lysates were immunoblotted for the indicated proteins.
