Inhibition of AP-1 activity suppresses the hyperosmotic secretion of PlGF from RPE cells. The levels of VEGF-A165 (A) and PlGF (B) proteins were determined with enzyme-linked immunosorbent assay (ELISA) in the media of cells cultured for 24 h under iso- (control) and hyperosmotic (+ 100 mM NaCl) conditions, and in the presence of the hypoxia mimetic CoCl2 (150 µM), respectively. The cells were cultured in the absence and presence of the inhibitor of AP-1 activity, SR11302 (SR; 5 µM), and the inhibitor of the FGF receptor kinase, PD173074 (500 nM), respectively. Each bar represents data obtained in four independent experiments using cell lines from different donors. Significant difference versus unstimulated control: *p<0.05.
