Activated GR signaling upregulates LEDGF/p75 protein expression in PCa cells. Cells were treated every 24 hours with 10 nM Dex or cortisol for up to 48 hours. Untreated (U) cells were used as controls. Whole cell lysates were prepared for Western blotting and probed with anti-LEDGF/p75 antibodies. Results from at least 3 independent experiments were quantified with ImageJ as described in the Methods section. Results reveal increased LEDGF/p75 expression in MDA-PCa-2b (A), 22Rv1 (B), and PC3 (C) cells and decreased LEDGF/p75 expression in DU145 (D) cells treated with glucocorticoids. Unpaired t-test statistical analysis revealed that LEDGF/p75 induction in MDA-PCa-2b and 22Rv1 cell lines achieved the highest statistical significance (*p < 0.05, **p < 0.01, ***p < 0.005).
