Activated GR signaling upregulates CLU protein expression in PCa cells. Cells were treated every 24 hours with 10 nM Dex for up to 48 hours. Untreated (U) cells were used as controls. Whole cell lysates were prepared for Western blotting and probed with anti-sCLU antibodies. Results from at least 3 independent experiments were quantified with ImageJ as described in the Methods section. Results reveal increased CLU expression in MDA-PCa-2b (A), 22Rv1 (B), and PC3 (C) cells treated with Dex. Increased CLU expression was not observed in DU145 (D) cells. Unpaired t-test statistical analysis revealed that increase in MDA-PCa-2b and 22Rv1 cell lines achieved the highest fold change compared to untreated controls (*p < 0.05, **p < 0.01). All blots represented within panels, although cropped, are derived from CLU and loading control images from the same gel. Full-length blots/gels are presented in Supplementary Figure S2.
