A) Primary XHIM patient CD4+ T-cells were electroporated with TALEN mRNA and transduced with an AAV6 codon-divergent CD40L cDNA donor. Expression of CD40L was measured by flow cytometry in resting T cells and after stimulation with anti-hCD3/anti-hCD28 microbeads. B) Average gene modification rates as measured by flow cytometry with and without stimulation. Data are presented as mean ± SD. n=8–10 biological replicates, 2 XHIM donors. C) CD40L expression trends by flow cytometry in XHIM T cells electroporated with TALEN and AAV donor and re-stimulated over time in culture.
