Cultured enteric neural crest‐derived cells and whole SC samples express common neuroglial markers. (A) Uncultured gut and SC tissues were analysed by qRT‐PCR to determine relative expression levels of the neurosphere markers. TuJ1 (pan neuronal marker), S100 (glia), Sox10 (progenitor/stem cells) and the neural crest marker p75. (B) Expression levels of the neurosphere markers were assessed in cultured ENS‐derived neurospheres and compared with expression in the SC. (C) Expression levels of specific neuronal subtypes were compared between uncultured gut and SC samples, including nNOS (NO), TPH1 (serotonin), GLS1 (glutamine) and ChAT (acetylcholine), and GAD (GABA) (D). The expression levels of these neuronal subtype markers were compared between ENS‐derived neurosphere cultures and uncultured SC samples. nNOS, TPH1, GLS1, GAD n = 3, ChAT n = 2. * ≤ 0.05, ** ≤ 0.005.
