Experimental features Lipid raft–enriched cell membranes were prepared by sucrose density gradient centrifugation in the presence of Triton X-100; purified membrane-associated glycosphingolipids were analyzed by MALDI-TOF MS using α-CHCA as the matrix.
cDNA was prepared from total cellular RNA, and the expression level of target genes was analyzed by real-time PCR using fluorescent-labeled hydrolysis probes.
