Mechanism of action of CTLA-4 inhibitors. Tumor cells develop B7-1/B7-2 to bind with CTLA- 4 on T cells, which prevents T cells from destroying the tumor cells (left). By blocking the ability of B7-1/B7-2 to bind to CTLA-4 with a CTLA-4 inhibitor, T cells are then able to kill the tumor cells (right). CTLA-4 = cytotoxic T-lymphocyte–associated antigen 4.
