Effect of Withaferin A (WA) on amyloid beta (Aβ) secretion. (A) Cells were treated with different dose of WA and the supernatants were collected 48 h after treatment. The supernatant collected were analyzed by Aβ1–40 ELISA which demonstrated that at an optimum dose of 2 μM WA, the levels of secreted Aβ40 showed significant decrease compared to control untreated samples. (B) The dosage of WA used for this experiment were also analyzed for the associated cellular toxicity. The cell toxicity assay showed that the lower doses of WA were not toxic to cells. Optimum dose of 2 μM, WA did not cause any loss in cell viability or toxicity in amyloid precursor protein (APP)-plasmid transfected SH-SY5Y (SH-APP) cells (*p ≤ 0.05; ***p ≤ 0.001; ****p ≤ 0.0001).
