Incorporation of de novo SAM for the MTase assay. De novo SAM was prepared using a reaction mixture containing 25 mM potassium phosphate (pH 7.5), 2 mM methionine, 1 mM ATP, 5 mM MgCl2 and his-tagged recombinant MetK (0.1 mg/ml). The reaction was incubated at 37°C without shaking for up to 1 h. HPLC chromatogram confirming SA MTase in the presence of de novo SAM.
