LexA-Grf10 overexpression triggers filamentation and requires a functional DNA-binding homeodomain. (A) Strains harboring the truncation and full-length LexA-Grf10 fusion constructs, as in Fig. 2, were grown on SC-Met or SC-Met−Ade medium for overexpression, as indicated. Yellow arrowheads indicate hyphal growth. Plates were incubated at 30°C, and photographs were taken at 72 h. (B) Left, strains expressing LexA-Grf10W83A-N86A (1, RAC300), LexA (2, RAC201), or LexA-Grf10 (3, RAC216) were grown overnight in YPD and normalized to an OD600 of 0.1, and 3 µl were spotted onto solid SC-Met medium. Plates were incubated at 30°C, and pictures were taken at 96 h. Right, a single colony of each strain was inoculated into SC-Met medium and incubated for 16 h at 30°C. Samples were examined by light microscopy at ×100 magnification. (C) Strains numbered as in panel B were spotted on solid medium, as indicated and as described in Fig. 2. Plates were incubated at 30°C, and pictures were taken at 48 h. At least three biological replicates were performed for each experiment.
