Inhibition of BRD4 decreases c-Myc and Bcl-2 mRNA transcription, but increases p21 mRNA transcription in prostate cancer lines. (A) DU145 and LNCAP cells were transfected with either shBRD4 or NC shRNA, and the relative mRNA transcription levels of c-Myc, Bcl-2 and p21 were analyzed by RT-qPCR. (B and C) DU145 cells were treated either with JQ1 (10, 20 or 40 µM) or DMSO, and LNCAP were treated with either JQ1 (100, 200 or 400 nM) or DMSO for 48 h; the relative mRNA transcription levels of c-Myc, Bcl-2 and p21 and were analyzed by RT-qPCR.
