(A) NRVMs were treated with sildenafil (PDE5i), and after 30 minutes cells were treated with sera from NF or SV patients for 72 hours. RNA was harvested for RT-qPCR and RNAseq. (B) Log2 relative mRNA expression of prototypical pathological cardiac genes (BNP, ANF, βMHC and αMHC) in NRVMs. For all groups, bar equals log2(mean±SEM), asterisk (*) denotes a significant difference between groups, one-way ANOVA, Holm-Sidak multiple comparisons (*p<0.05, **p<0.01, ***p<0.001, ****p<0.0001), n=5 NF, n=16 SV, n=4 NF+PDE5i, n=14 SV+PDE5i in a total of 7 NRVM preparations.
