Changes in Cl− flux did not affect AQP1-AE1 interaction in erythrocyte IOVs. In this set of experiments, IOVs singly labeled with BRIC170-Alexa Fluor 488 conjugate, or colabeled with anti-AQP1-Alexa Fluor 568, were suspended in half-Cl− HBSS for FLIM measurements. Despite the reduced Cl−, fluorescence lifetimes of FRET donor Eg decreased significantly in the presence of the acceptor Qr.
