Vanillate demethylation requires specific accessory homologs. Vanillate degradation involves the transfer of a methyl group from vanillate to tetrahydrofolate, which is then oxidized to formate. Two homologs each of metF and folD are present in the strain DSM12444 genome. Pooled fitness measurements readily identify the specific homologs that are required for vanillate demethylation. Error bars show one standard deviation, calculated from three biological replicates.
