The reduction of Treg cells in ongoing PCM of DEREG mice increases the number of pulmonary Th1 and Th17 cells but reduces the presence of Th2 lymphocytes. The phenotypic analysis of lung infiltrating lymphocytes from DT and PBS (control) treated DEREG mice was performed at weeks 6 and 10 of P. brasiliensis infection. The lung cells were obtained as described in Material and Methods and labeled with antibodies conjugated to different fluorochromes. The lung infiltrating leukocytes were gated by FSC/SSC analysis. The cells were gated for CD4+ (A) or CD8 (B) expression and then for intracellular expression of IFN-γ, IL-4 and IL-17. One hundred thousand cells were acquired on FACS CANTO II and subsequently analyzed by FlowJo software. Data are expressed as means ± SEM of three independent experiments using 5 mice per group (*p < 0.05, **p < 0.01 and ***p < 0.001).
