BJAB RRV cells were induced with 100 nM trichostatin-A (TSA) following electroporation with antisense oligonucleotides complementary to either GFP mRNA (control KD) or RRV PAN RNA (Oligo 493 and Oligo 539). (A) Total RNA was processed 40 h after lytic induction. PAN RNA levels were quantified by Northern blot. (B) Seven days after induction, encapsulated viral DNA released into the media was harvested and quantified by qPCR; values were normalized to a control plasmid added at the onset of purification. (C) Seven days after induction, intracellular DNA was harvested from the cells and the level of viral DNA relative to host DNA was determined by qPCR. Data are the average of three biological replicates and error bars represent standard deviations of the mean.
