Pan-NOTCH/γ-secretase inhibitor BMS-906024 delays H1299 multicellular spheroid specific growth in combination treatments. SSGD of multicellular NCI-H1299 spheroids were treated with 2.5 nM paclitaxel (A) or 0.4 μM crizotinib (B) and/or 1 μM BMS-906024 and/or 2 Gy radiation, is shown. Statistical significance of the comparison between single-agent chemotherapy: paclitaxel (A) or crizotinib (B), vs. chemotherapy plus BMS-906024 (left), or chemoradiation vs. chemoradiation plus BMS-906024 (right) is shown. Spheroid specific spheroid growth delay (SSGD) was calculated as T4xSVtreatment – T4xSVcontrol and corrected for the doubling time of the control group. A minimum of three independent biological replicates with 12 spheroids/condition/experiment were tested. Mean and standard error of the mean are plotted. p-value < 0.001 (***) and < 0.050 (*).
