Effects of SE1 on activation and expression of MMP-9 and MMP-2 in HT1080 cells. (a) Gelatin zymography for the determination of MMP-2 and MMP-9 activities in SE1-treated HT1080 cells. HT1080 cells treated with SE1 (10, 20, 50, and 100 μM) for 1 h and stimulated by PMA (10 ng mL−1) for 72 h. Gelatinolytic activities of MMP-2 and MMP-9 in conditioned media were detected by electrophoresis of soluble protein on a gelatin containing 10% polyacrylamide gel. Untreated control was used as a loading control. (b) Expression of MMP-2 and MMP-9 in cell lysates was detected using western blot analysis. β-actin was used as a loading control. HT1080 cells treated with SE1 (10, 20, 50, and 100 μM) for 1 h and stimulated by PMA (10 ng mL−1) for 24 h. The relative amount of MMP-2 and MMP-9 was quantified by densitometry measurement (ImageJ).
