NF-κB activation by reptilian TLR15. HEK293 cells transiently transfected with an NF-κB luciferase reporter plasmid and empty vector, chicken (gaga), anolis (anca), crocodile (cpro), alligator (almi) TLR15 or the gagaTLR2B and gagaTLR1A plasmids were stimulated (5 h) with Proteinase K (100 ng/mL), Pam3CSK4 (100 ng/mL), FSL-1 (100 ng/mL), 10 μL of Chrysosporium anamorph of Nannizziopsis vriesii (CANV) sterile culture supernatant or 10 μL of CANV supernatant pre-treated (30 min) with 1 mM PMSF. Values are the mean ± SEM fold increase of NF-κB activity, represented by luciferase activity in Relative Light Units (RLU), in stimulated cells over unstimulated control cells from three independent experiments performed in duplicate.
