Detection of FLAG SOX9 expression in rAAV-transduced human bone-marrow aspirates seeded in 3D-woven PCL scaffolds. Aspirates were transduced with rAAV-lacZ or rAAV-FLAG-hsox9 or left untreated and seeded in the scaffolds in fibrinogen/thrombin using chondrogenic medium, as described in Fig. 1 and in the Methods. The samples were processed after 21 days to detect SOX9 (magnification 10 × ) and FLAG expression (inserts: magnification 20 × ) by immunohistochemistry (all representative data). Scale bars: 200 mm.
