The bicyclic peptides 1b and 3b and monocyclic peptide 6c (100 μM) were incubated with trypsin (0.05 μg/mL) at 37 ºC. After 1 hour, less than 5% of 6c remained, while 45% of 3b and 70% of 1b remained, demonstrating that bicyclization improves proteolytic stability relative to monocyclization.
