Confluent NHEKs were incubated in a high calcium medium (1.3 mM) for 16 h with or without tunicamycin pretreatment (100 ng/ml). Cycloheximide (30 μg/ml) was added, and the cells were harvested at 0, 1, 2, and 4 h after the cycloheximide treatment. The dashed lines indicate half the amounts of Dsg1 and 3 found at 0 h; n=3. Data are depicted as means ± S.E.M. Statistical analysis was performed using repeated-measure ANOVA with Tukey’s post hoc test; *P<0.05. Abbreviation: CHX, cycloheximide.
