Schematic representation for the generation of rZIKV: Mammalian Vero cells are transiently transfected with infectious RNA transcribed in vitro from a full-length cDNA clone (top), full-length infectious cDNA clones (left) or infectious subgenomic amplicons (right) (see also Figure 2). After 3–4 days post-transfection, when cytopathic effect is observed, rZIKVs present in tissue-culture supernatants can be recover for titration or viral amplification in mammalian (Vero, left) or insect (C6/36, right) cells.
