Plot of the relative yields of the 17-mer transcripts from templates with different sequences around Ds, with natural NTPs (1 mM), in the presence of each modified-PaTP or PnTP (1 mM). The relative yield of each full-length transcript was determined by comparison to the yield of the native transcript from the template consisting of the natural bases only (3′-N₁DsN₂-5′ = 3′-CAT-5′ for ′-CDsC-5′, 3′-CDsT-5′, 3′-TDsC-5′; 3′-N₁DsN₂-5′ = 3′-GAG-5′ for ′-GDsG-5′). The values were averaged from three to four data sets.
