Elution profile of the Sephadex^® G-50 affinity chromatography. Approximately 10 mL of the crude extract was applied to the column (6.5 × 1.8 cm) which was equilibrated with 150 mM NaCl plus 5 mM CaCl₂ and 5 mM MnCl₂. The lectin was eluted with 100 mM D-glucose in the equilibrium solution described earlier at a flow rate of 1 mL/min. Fractions of approximately 3 mL were collected and monitored for protein content by measuring the absorbance at 280 nm.
