Uprolides compounds inhibit the production of TNF (A) and IL-6 (B) induced by LPS in macrophages. Peritoneal macrophages from C57B1/6 mice were treated with different concentrations of compounds 1, 2 or 3 (uprolides N, O or P) 1 h before stimulation with 100 μg/mL of LPS. The supernatants were harvested after 6 h, and cytokine concentrations were determined by ELISA. Results are represented as Mean ± S.E.M. from two independent experiments performed in duplicate. **, p < 0.01, ***, p < 0.001 relative to the LPS stimulus alone. (C) The cell viability was assessed using a MTT assay after collecting the supernatant.
