4Mu Decreased CD133+ CSC Capability to Growth and Disseminate
(A) C3Hj/He mice (n = 4–5 per group) were inoculated (i.v.) with (1) 1 × 105 CD133− cells; (2) 1 × 105 CD133− cells plus 4Mu; (3) 1 × 105 CD133+ cells; or (4) 1 × 105 CD133+ cells plus 4Mu. Animal survival was analyzed. ***p < 0.001, Kaplan-Meier, log-rank test. (B) Tissue sections of mice receiving CD133+ cells showed more metastatic HCC nodules within the lungs in comparison with mice receiving CD133+ plus 4Mu or CD133− cells. (C) CSC gene expression profiling by PCR array. (D) Real-time PCR confirmed downregulation of Nanog, Wnt, Sox, and Zeb1 expression in tumor samples of 4Mu+AdIL-12-treated mice compared to control mice. *p < 0.05; **p < 0.001; and ****p < 0.0001 versus saline (ANOVA and Tukey's post test). Data are expressed as the mean ± SEM.
