Fig. 2.
MOPr activation in N2AMOPr cells causes significant upregulation of RTP4 mRNA levels. (A) N2AMOPr cells (3.2 × 104 cells/well) were treated without or with DAMGO (10 µM) for 20 minutes (first DAMGO) and the Gαi response was measured using the CellKey System. After 24 hours, cells were given a second treatment with DAMGO (10 µM) (second DAMGO) followed by measurement of the Gαi response. Data are the mean ± S.D. n = 8–9. **P < 0.01 vs. control; ##P < 0.01 vs. first DAMGO, Tukey’s test. (B) N2AMOPr cells (2 × 104 cells/well) were treated with DAMGO (10 µM), dopamine (10 µg/ml), or serotonin (10 µg/ml) for 24 hours. RT-qPCR was performed with specific primers against RTP1, 2, 3, 4, and GAPDH. Data are the mean ± S.D. n = 7–45. **P < 0.01; *P < 0.05 vs. control, one-way multivariate ANOVA and Bonferroni test.
