E007 cells stimulate the TSCM differentiate rapidly and the daughter effectors produce cytokines. The prepared TSCM were stimulated with E007, E001, and α-CD3/CD28, respectively. a, b The TSCM differentiated into effector T cells upon the stimulation with E007. CD3+ CD8+ T cells expressing CD45RA and CD62L (TSCM cells) were detected at indicated hours after stimulation. Representative FCM plots were shown (a). Data are represented as mean ± SD of four individual experiments (b). cf Effector T cells differentiated from the TSCM produced cytokines. Intracellular IL-2, TNF-α, and IFN-γ production of the T cell subsets were detected. Gating by CD62L, the T cells were divided into CD62L+ (TSCM and TCM) and CD62L- (TEM and TEF) cells, the TEM and TEF subsets showed the cytokine positively stained cells. Representative FCM plots (the percentages indicated the cytokine producing cells in CD62L+ and CD62L- cells, respectively) (c) and the frequencies of IFN-γ (d), TNF-α (e) and IL-2 (f) positive cells in corresponding CD8+ T cell subsets were shown. Data are represented as mean ± SD of four individual experiments (**p < 0.01; ***p < 0.001, and NS, p > 0.05)
