EP2, DP1 and PPARγ are critical for mediating the effects of PGE2, PGD2 and 15d-PGJ2 on the regulation of APH-1α/1β and PS1 expression in n2a cells. (A) n2a cells were treated with PGE2 (10 μM) in the absence or presence of AH6809 (3 μM) for 48 h. (B) In select experiments, n2a cells were treated with PGE2 (10 μM) in the absence or presence of siRNA specific for EP2. (C) In select experiments, n2a cells were treated with PGD2 (1 μM) in the absence or presence of siRNA specific for DP1. (D) In distinct experiments, n2a cells were treated with 15d-PGJ2 (500 nM) in the absence or presence of the PPARγ antagonist GW9662 (1 μM) for 48 h. The levels of APH-1α/1β and PS1 were determined by qRT-PCR and western blots, respectively.
