Disruption of de novo pyrimidine biosynthesis results in reduction of intracellular UMP and UTP, and supplementation of uracil to the medium restores the levels of UMP and UTP in P. aeruginosa. P. aeruginosa strains were grown on PIA at 37°C for 24 h. The cells of each type in triplicate were harvested, washed, and resuspended in PBS. The cell-free lysates were used for measurement of intracellular concentration of UMP (A) and UTP (B) measured using LC-MS/MS. Two-way ANOVA was used with statistical significance at P < 0.01 (*, P < 0.01; **, P < 0.001; ***, P < 0.0001).
