Test process of polymerases by the example of Probe/ROX qPCR Master Mix. 1.1 prfA and ΔprfA assays were initially carried out under the thermal profile of the established prfA assay (MgCl₂ concentration as recommended by the supplier). 1.2 Afterwards, assays were adapted for amplification of all DNA concentrations of the calibration curve (1.58 × 10¹-1.58 × 10⁶). 1.3 Finally, assays were validated using Poisson and PCR-Stop analysis (modified after [21]).
