Rapalogs form a complex with FKBP12 that inhibits mTORC1 activity, S6K activation but only partially reduces 4EBP signaling. Rapalogs also prevent the feedback inhibition on the PI3K/Akt signaling, thus converging toward Akt activation and cancer cell survival. Prolonged exposure to rapalogs partially inhibits mTORC2 assembly and mTORC2 functions on Akt signaling.
