Overexpression of p62 attenuated escin-induced cell apoptosis. (A) The effects of escin on cell viability after p62 overexpression. Cells were transiently transfected with FLAG-NC and FLAG-p62 for 36 h and then treated with 60 μg/mL escin for 12 h. Cell viability was assessed by the CCK8 assay. (B) Analysis of apoptosis with FACS in HCT116 cells. Cells were treated as described above, and apoptosis was analyzed with FACS. Right: quantitative analysis of apoptosis after p62 overexpression with or without treatment of escin is shown. (C–D) Expression of apoptosis related proteins. Cells were treated as described above, and apoptosis-related proteins were detected by Western blotting (C).
