Figure 1. — Total membrane fractionation of BY2 cells expressing HA-tagged Lr34res protein. A, fractions ranging from 7.4 to 48.6% (w/v) sucrose were harvested after ultracentrifugation and tested for the presence of HA-Lr34res and marker proteins of the PM (H⁺-ATPase), the tonoplast (V-ATPase), thylakoid membranes (PsbA), and Golgi membranes (Arf1) by Western blotting using specific antibodies. Fractionation was repeated twice with the same outcome of HA-Lr34res distribution. B, quantification of signal intensities after blot development. For each protein, values are relative to the fraction with the strongest signal, which was set to 100%.
