After pretreatment in the presence or absence of NF-κB inhibitors BAY 11–7082 (10 μM; a–d) and TPCA-1 (10 μM, e–h) for 1 h, canine melanoma cells were incubated with or without 100 pM IL-1β for 15 min, and then the phosphorylation levels of p65 and p105 were determined by immunoblotting. For the immunoblotting, cell lysate (10 μg protein) was used. Representative results (a, c, e, and g) and the relative density of p-p65 (b and f) or p-p105 (d and h) expression compared to the results without the inhibitor and IL-1β are illustrated. Values are expressed as the mean ± S.E. of three independent experiments. *P < 0.05.
