P. falciparum had1 mutant strains resistant to fosmidomycin (1a) are also resistant to MEPicides. Dose-dependent growth inhibition was determined for P. falciparum strains treated with inhibitors as previously described.³⁵ The had1 mutation results in higher levels of the DXR substrate DOXP, and mutants are resistant to DXR inhibition as indicated by a shift in the IC₅₀ curve (had1; open shapes, black line) when compared to WT P. falciparum (3D7; closed shapes, gray line). Sensitivity was restored if a WT copy of had1 was supplied in the mutant strain (had1 + HAD1-GFP; closed shapes, black line). Data are representative of at least three independent biological replicates performed in duplicate.
