Lead compound and synthetic approach. (A) The lead 1 is shown with the Areas A, B and C highlighted. These areas are the focus of diversification in analog synthesis to explore structure-function relationships in the lead series. (B) Retrosynthesis of the 5-amino-1-(carbamoylmethyl)-1H-1,2,3-triazole-4-carboxamides is shown, with the core of Area B formed via a cycloaddition of azide 15 and nitrile 16.
