lncRNA‐MALAT1 is induced by hypoxia and regulated by HIF‐1α in human endometrial stromal cells. A, Human endometrial stromal cells were incubated under hypoxic conditions for 0, 4, 8, 16, and 24 h. lncRNA‐MALAT1 levels was examined by qRT‐PCR. (B‐C) Human endometrial stromal cells were transfected with empty vector or HIF‐1α expression plasmid before culture under normoxia for 24 h. HIF‐1α protein expression levels was examined by Western blot. D, Human endometrial stromal cells were transfected with empty vector or HIF‐1α expression plasmid before culture under normoxia for 24 h, and lncRNA‐MALAT1 levels was examined by qRT‐PCR. (E‐F) Human endometrial stromal cells were transfected with control siRNA or HIF‐1α specific siRNA before culture under normoxia for 24 h. HIF‐1α protein expression levels was examined by Western blot. (G‐H) Human endometrial stromal cells were transfected with control siRNA or HIF‐1α specific siRNA before culture under normoxia or hypoxia conditions for 24 h, and lncRNA‐MALAT1 levels were examined by qRT‐PCR. The protein expression levels were quantified by Image J software and normalized to GAPDH protein levels. Quantitative results are expressed as the means ± SD of at least three independent experiments (*< 0.05; **< 0.01)
