Internalization of surface-expressed CD103 in human MoDCs. Human MoDCs were labeled with an anti-CD103 FITC (mouse IgG1), washed, and chilled with ice-cold buffer. Time zero samples were left on ice, and internalization was initiated in the remaining samples by resuspending the samples in 37°C media and incubating them at that temperature for the indicated times. Following incubation, cells were harvested with cold buffer and then left on ice until all samples were collected. Anti-CD103 antibody remaining on the surface of the DCs was detected with a secondary anti-mouse IgG1 antibody.
